Esperion Therapeutics, Inc. The program centers on Esperion’s next-generation allosteric ACLY inhibitors, with lead candidate ESP-1336 showing encouraging preclinical results. Studies demonstrated reduced liver inflammation, injury, and fibrosis across multiple PSC models. The company is aiming to leverage its ACLY biology expertise—previously applied in cardiovascular drug development—to expand into hepatic diseases, where it estimates a market opportunity exceeding $1 billion.

CEO Sheldon Koenig emphasized the unmet need in PSC and the company’s commitment to addressing it, stating, “Our lead candidates represent a natural extension of our metabolic franchise.”
